Abstract

Breast cancer has monoclonal origin in stem-cells or progenitor cells by means of genetic (dependent on DNA damage) and epigenetic mechanisms (regardless of modifications in the DNA sequence). The process of suppressor genes inactivation necessary for mammary carcinogenesis initiation in sporadic (non-familial) or hereditary (familial) cancers, considering the two-hit model in the homologous chromosomes (Knudson theory) is herein presented. Genetic mutations are discussed as well as the epigenetic alterations that cause gene silencing, as DNA methylation, histones modifications and microRNA.
